SMART Chromatography™ Column for His-tag Purification Using Metal-Free NTA Resin
SMART Chromatography™ NTA (metal-free) from emp BIOTECH is a ready-to-use column packed with ZetaCell NTA resin—a hydrophilic, cross-linked agarose medium functionalized with uncharged nitrilotriacetic acid (NTA). Designed for use with SMART Chromatography™, the column supports purification of His-tagged recombinant proteins when charged with divalent metal ions such as Ni²⁺, Co²⁺, Cu²⁺, or Zn²⁺, offering flexible metal coordination based on protein-specific binding preferences.
The 250 µm agarose beads provide a low-backpressure, high-flow matrix optimized for direct processing of non-clarified cell lysates. Proteins are captured via selective affinity to chelated metals and eluted under native or denaturing conditions using imidazole, histidine, or low-pH buffers. The column supports linear scalability from lab-scale testing to full manufacturing using SMART Radial Flow Chromatography.
